Doctor, V. M. (University of Texas, Houston). Microbiological assay for thymine using a mutant of Streptococcus faecalis. J. Bacteriol. 84:17-22. 1962.-A rapid microbiological assay procedure for the determination of thymine was devised. The test organism, Streptococcus faecalis Akt, a mutant of S. faecalis ATCC 8043, gives one-half the maximal growth response to 1.6 mug of thymine (assay range 0.5 to 2.5 mug) or to a molar equivalent (3.2 mug) of thymidine. Under these experimental conditions, 80 mug of 5-methylcytosine or 150 mug of thymidylic acid replace the thymine or thymidine requirement of the test organism. Addition of a mixture of adenine, guanine, and xanthine at a level of 40 mug/ml of each or higher to the basal medium, or addition of a mixture of deoxyadenosine and deoxyguanosine at a level of 80 mug/ml of each or higher to the basal medium, inhibited the thymine or thymidine growth response of the organism. Thymine activities of rat and chick tissues were determined by this procedure. The data on the recovery of added thymine to tissues gave 84 to 103% recovery. Thymus, spleen, small intestine, and ventral prostate were higher in thymine activities as compared with other tissues.

The utilization of dihydrothymine by Streptococcus faecalis in the absence of folic acid and thymine.
Stokes (1944) reported the replaceability of folic acid by thymine for Streptococcus lactis strain R as well as other folic-acid-requiring streptococci, and Fink et al. (1954) noted that high concentrations of recrystallized dihydrothymine supported growth of Streptococcus faecalis (ATCC 8043) deficient in both folic acid and thymine. The question arose, however, as to whether the latter respon...

A microbiological assay has been developed for chromomycin A(3), an antitumor antibiotic showing promise in human trials. The assay bacterium is a derived strain of Streptococcus faecalis resistant to methotrexate. Studies with mice revealed that relatively high concentrations of this antibiotic were maintained in the blood, kidneys, and liver of mice after a single-dose intraperitoneal injecti...

The Autoturb System, a semiautomated system for photometric bioassay, was used to determine cefazolin content. Suitable conditions for the assay using Streptococcus faecalis ATCC 10541 as the indicator organism included a medium pH of 6.0 to 7.0 and an incubation time of 3 to 3.5 h at 36 C. Multiple independent assays of samples from a common batch showed the test to be highly reproducible. Acc...

BACKGROUND Streptococcus mutans, Candida albicans, and Enterococcus faecalis are the three oral microorganisms most commonly implicated in the causation of oral infections. All these oral microorganisms have shown resistant to routinely used antimicrobials. There is a need for an antimicrobial agent which is effective, safe, and economical. Zingiber officinale, commonly known as ginger is one s...
